Providing an interface material to prevent metal contact of the heatsink/water block with the PCB is the responsibility of the cooler manufacturer. Any product that does not include this type of insulator is junk at best and should be avoided.

Even auto-routing is constraint by specifications. Also at this level of complexity auto-routing is not the final step. Human tweaking is needed for the finished design.

Yes, two of those fans can be connected to the same header without issue. Noctua states the included splitter will properly control both connected fans.
