Due to convenience and laziness, when I bought an extra SATA drive for a PC, I didn't reformat the EIDE hard drive and make the SATA the main drive to boot the operating system from.

Would I notice a significant improvement in system performance if I reformat and make the SATA the primary drive?

The most I'd expect is a faster startup of Windows, but I could even be wrong there.

The system is a Pentium D 820, 1GB DDR2-533, soon to be Q6600, 3GB DDR2-533, if that's relevant at all.

Thanks!

You might. You might not.
What are the specs of the drives?

Also, you don't have to reformat if you want to use the SATA drive as your OS drive. You can image the IDE to SATA using drive imaging program.. (Acronis TrueImage, Norton Ghost are popular)

just make sure your controller card / motherboard can boot from sata
and that you have a way to get into windows successfully on the sata drive before formatting / deleting the IDE drive.
