What exactly is PCI-E?

Is a video card that is PCI-express, plug into a regular PCI-Slot? Or do u need a more high end motherboard?

Yes, PCI-Express is a specifically designed slot for PCI-E hardware (as of now mainly used for PCI-E video cards). It is jsut a slot that transfer data and processes it much faster than both an AGP and PCI slot. Most high-end motherboards are equipped with such a slot.

PCI-Express card goes only in PCI-E slot, nowhere else. It is more advanced slot than AGP, so you can't stick AGP card in PCI-E slot or vice versa.
 
Not nessisarily High End Mobos, They can be found for quite cheap ($90-$200).

Also, Most Intel PCI-E Mobos are socket 775, while most amd PCI-E Mobos are socket 939 (there is a 754 one out, but if you are upgrading I suggest 939)
