I've recently switched to Vista due to the new system build and apparently many people are having a similar problem as I do. I can't use my mic - the "Microphone" input in Vista is checked but there is no jack information. The other "Microphone" device with the jack information is grayed out. The card I'm using is Asus Xonar DX and I'm plugging the mic into the only input jack it has. I played with the Asus software a bit, and in the mixer options I have the mic checked. I have the latest Asus drivers installed for the soundcard.

I run Vista x64, and just recently installed the Xonar DX card. The microphone does work for me. I had to right click the microphone setting in Vista and make it the default input.

Did you remove any previous sound card files that might be left over? I don't have any other microphone devices displayed in there, just the one, along with a "line in" and 3 other grayed out options (Aux, Stereo Mix, Wave).
