I currently have a BFG GTX280 card, I'm looking to upgrade to 5xx series of card soon. My question is is what happens if I try to SLI the 2 cards? Will it lower the performance? Can I set the 280 to handle just the Physx?

You will not be able to SLI those cards. You will however, be able to set the GTX280 as your secondary card (meaning the 5xx series card must be the main card) for PhysX. That will be a VERY powerful PhysX card. However, if you get a GTX570 or 580 you probably won't need a dedicated PhysX card.
