One is with PSI Express and the Other isn't, what exactly does PCI Express do?

I'm getting this card for future plans do run SLI. I have a MSI K8N Neo4 Plat/Athlon 64 3200 Winchester/G.Skill DDR PC3200 1GB (512 x 2)/OCZ Powerstream 420w PSU.
 
PCI-Express is the interface for the video card. In order to do SLI you need two of the same PCI-Express video cards.

AGP won't fit in a PCI-Express slot, which your motherboard has 2 of for SLI.
