I need some help understanding how multi-pathing functions.

My OI NAS/SAN has 4 GBe nics w/ 2x (4x Raid 5 1TB drive)
one of my ESXi devices has two GBe nics
my other has four GBe nics

I was originally running iSCSI on my two ESXi devices but was mounting via one target. After some instability after upgrading to 4.1 I switched everything over to NFS. I am still not sure if that was the correct option but its been stable since the switch. The question now is I finally got a switch that supports enough ports to make connecting with all the nics a viable option.

Please help me understand what the correct path is and how I should accomplish the fastest throughput between OI and my ESXi Devices.

You don't multi path with NFS. You can export multiple datastores over different IPs and connect to those over different vmkernel interfaces...but no way to multi path to a single NFS server. Not yet...

So how does it function via iSCSI?

Is it more beneficial to run all my virtual on different sections of the hard drives with a different NFS share per virtual or target?

Also, look at your NIC utilization for storage. Are you hitting 100%? If not..multipathing doesn't buy you anything.
