Will it make that much of a difference in temps between these two loops?

Res->Pump->rad->block->res

and

Res->Pump->block->rad->res
 
Zero noticable difference in a closed loop as long as you have res > pump, but even that is debatable...

Edit: Forgot to mention, basically keep your tubing as short as possible with no elbows with res feeding your pump other than that you wont notice any performance difference in order
 
Loop order doesn't matter at all. The only reason the res should be before the pump is because it helps with bleeding.

Elbows impart a temperature difference of about 0.1c each.
