I just recently purchased a new SSD drive for cloning Windows 10 as a back up. Is there any reason to unplug it or is it okay to leaves plugged in and powered up.

If it's the only backup, I'd disconnect/remove. If you have a power supply failure, for example, you could lose one or all of the drives.

On a side note, I keep my backup drive connected, but it's in a different PC. ie: I backup from one PC to another.

You should have multiple backups. A live attached drive is subject to hardware failure, malware (ransomware, for example) or accidental erasure. Have a live, connected backup for immediate backups on a schedule, and a disconnected backup (which by its very nature you will most often NOT be updating as often) to mitigate the chances of the problems listed and kept in a separate location.
