I have a windows Server 2003 Machine that i use to host all my media for my house, and a few small websites. It is connected to a dlink router through two NICs. I wanted to know if it were possible to designate 1 interface for external traffic, and one for internal traffic?
 
Not unless you have two separate networks - one external and one internal. You might not want to do that anyway since it could pose a security risk vs putting the server in a DMZ.
 
Give each interface a unique IP. Configure the websites to only respond on one IP. Configure the media clients to use the other.

Just a note: as far a bandwidth is concerned, if both NICs are connected to the same cheapo switch, you are going to see the same contention for traffic as you would using a single NIC.
