I would like to partition my 250GB hard drive into smaller partitions to increase performance, and make things more organized. How do i go about doing this? Do I have to format it in DOS, then after that use fdisk to create the partitions?

I dont believe you will reap any performance gains from partitioning a drive. That opinion stated, just install the drive and right click my computer > manage > disk management. From there you can slice and dice the drive as much as you want.

If you want to do this on a fresh install, just create your os partition as whatever size you want from the setup. Install windows and see above. You can just as easily create all partitions from the setup ("C" when prompted then enter the size in MB)but you will only be able to format one of them from there.

Any gains you get from intensifying locality by partioning the drive will be only a few percent, and will be offset by artifically lengthening seeks between partitions.

The case for organization is a little better, and having spearate file systems does make your data a little less vulnerable to fragmentation, corruption, and other file system level problems.

You can create four primary partitions or three primary and one extended partition on a basic disk. The extended partition may be further subdivided into logical drives as needed. Dynamic disk volumes are more flexible in terms of layout, but can complicate disaster recovery.
