It is a total Revamp of the way you put cards into your computer. Take everything you know about current PCI bus architectures, and turn it around with the exception of that they both use slots. Here is a great explanation of the information your are probably looking for.

Is it likely that the next generation video cards will be using PCI Express?

Yes, PCI-E will require a new board Yes, R420 and NV40 will be AGP cards but after that, cards will switch over to PCI-E. However, there will probably be some AGP graphics boards in circulation and the production of AGP cards will most likely continue for some time. (I'm just speculating on some of this stuff, but I feel I might be somewhat accurate.)
 
cant wait until PCI-E goes into full swing ..lets see if it lives up to the hype
 
I doubt if it will change anything in the near future with video card, but being able to actualy use Gigabit ethernet will kill ass. And it's also time for realy realy realy powerful sound cards :D
