I'm a little confused the board has 3 screws or are these the standoffs? I do have one just one screw that came with the motherboard which fits in I couldn't find a simlar ASROCK board that looks the same for a video tutorial.

You just use the hole where the SSD ends, the different holes are for different sizes.

I know that much but not sure if the screw in the MB is the stand off. The two pictured to the left so if it goes over the ssd or under.

You need to move the standoff to the appropriate location for your M.2 drive. The standoffs I see in the pictures will unscrew so you can relocate to the proper position then use the screw to secure the drive.
The screw is not the standoff.

The silver/grey is nut is the standoff, the screw comes on a plastic bag on your mobo box, here is my AsRock Z370 Taichi with EK NVME heatsink on 970pro.
