If I moisten the bi flanges (or tri flanges) of my er-6is and fit them nicely up in the ear canal getting a good seal I really can't hear anything but music.
I usually reinsert them a few minutes after the initial insertion to get them sealed well rather than wetting them first.
5.1 headphones have been considered the snake oil of headphones. You'd have to ask someone who has a set of 5.1 and a set of stereo headphones around the same value to back that up.
The Audio Technicas don't require an amp, that is a nice feature of them. They will sound better with an amp but...
Do a search on this forum for 5.1 headphones and you'll turn up mixed opinions. The general opinion is that 5.1 headphones are not worth your time/money.
You can get a much better set of headphones for that budget. I have the Audio Technica ATH-a900's so I believe I can vouch for the sound...

The a900's get a little warm but I don't feel any pressure from the pads on top. I'm wearing them right now and I don't even have sound going through them, I just wear them to drown out fan noise sometimes :)

Technicalities aside, why not test each of your theories by playing a game/movie and finding out what works best for your speakers/ears/drivers/receivers/soundcards/earwax. I don't think anyone should follow a spec when they can clearly hear sounds coming from somewhere they shouldn't be coming...
You should be fine if you've only done it once. Repeatedly using isopropanol and/or ethanol will deteriorate the plastic and make it turn yellow and brittle. Don't even think about using Windex.

Yes it has a headphone out but you're still going through all of the circuitry that the speaker manufacturer made, if we're talking about multimedia speakers here (ie. logitech, altec lansing). If you're just using a receiver, usually that's just fine but multimedia speaker headphone jacks...
